核酸適配體-癌癥研究的分子工具 譚蔚泓1,2 1分子科學(xué)與分子醫(yī)學(xué)實(shí)驗(yàn)室,化學(xué)生物傳感與計(jì)量學(xué)國(guó)家重點(diǎn)實(shí)驗(yàn)室(湖南大學(xué)),湖南大學(xué)化學(xué)化工學(xué)院,湖南大學(xué)生物學(xué)院,湖南大學(xué)分子工程與診療學(xué)協(xié)同創(chuàng)新中心,湖南,長(zhǎng)沙,410082 *Email: tan@chem.ufl.edu 疾病標(biāo)志物分子探針的發(fā)展有助于更全面的了解疾病的分子機(jī)理,但是此類分子工具在醫(yī)藥的臨床應(yīng)用研究中一直非常匱乏。最近,科學(xué)家們發(fā)現(xiàn)了一種新型的分子探針,稱之為核酸適配體。它是由DNA/RNA單鏈構(gòu)成的核酸分子,可特異性地識(shí)別單個(gè)蛋白、小分子;诖耍覀兪讋(chuàng)了以完整的細(xì)胞為篩選靶標(biāo)的核酸適體細(xì)胞篩選新方法(Cell-SELEX),該方法簡(jiǎn)單、快速、并具有可重復(fù)性。通過(guò)這種篩選方法,我們得到了多種疾病的核酸適配體探針,并將之用于生物醫(yī)學(xué)的前沿研究中,包括癌癥的超靈敏檢測(cè)、分子成像、藥物的靶向輸送,尤其是癌癥生物標(biāo)志物的發(fā)現(xiàn)。這些嶄新的分子工具為分子醫(yī)學(xué)和轉(zhuǎn)化醫(yī)學(xué)的更深入發(fā)展建立了一個(gè)革命性的科學(xué)平臺(tái)。本報(bào)告將介紹我們?cè)诤怂徇m體研究領(lǐng)域的最新研究進(jìn)展,特別是在分子工程、癌癥的生物標(biāo)志物,納米生物醫(yī)學(xué)及診療學(xué)中的應(yīng)用。 關(guān)鍵詞:核酸適配體;納米生物醫(yī)學(xué);生物醫(yī)學(xué);Cell-SELEX;分子工具;癌癥研究 參考文獻(xiàn) [1]Tan,W.; Fang, X.Aptamers Selected by Cell-SELEXfor Theranostics, Springer, March, 2015, 352 pp
The Molecular Foundation of Cancer: A Chemical Biology Approach Weihong Tan1,2 1 State Molecular Science and Biomedicine Laboratory, State Key Laboratoryof Chemo/Biosensing and Chemometrics, College of Chemistry and Chemical Engineering, College of Biology, and Collaborative Innovation Center for Molecular Engineering and Theronastics, Hunan University, Changsha 410082, China A full understanding of the molecular basis of diseases depends on the development of molecular probes able to recognize disease targets of interest. Until very recently, such tools have been absent from the clinical practice of medicine. The newest molecular probe, and one that holds most promise, is a new class of designer nucleic acids, termed aptamers, which are single-stranded DNA/RNA able to recognize specific targets, such as single proteins and even small molecules. Recently, we applied a simple, fast and reproducible cell-based aptamer selection strategy called Cell-SELEX which uses whole, intact cells as the target for aptamer selection. This selection processthen generates multiple aptamers for the specific recognition of biological cells, but without the need for prior knowledge about the signature of target cell-surface molecules. The selected aptamers have dissociation constants in the nanomolar to picomolar range. Thus far, we have selected aptamer probes for many different diseases, and used them to carry out studies at the vanguard of biomedical science, including ultrasensitive detection of tumors, molecular imaging, targeted drug delivery, and, most critically, cancer biomarker discovery. Taken together, these molecular level tools form a solid scientific platform from which to pursue advanced studies in molecular medicine. We will report our most recent progress in this exciting research area, especially in molecular engineering and molecular elucidation of cancer biomarkers and theranostics. |

譚蔚泓 | 譚蔚泓,中國(guó)科學(xué)院院士,1993年美國(guó)密西根大學(xué)博士,現(xiàn)任化學(xué)生物傳感與計(jì)量學(xué)國(guó)家重點(diǎn)實(shí)驗(yàn)室(湖南大學(xué))主任,湖南大學(xué)化學(xué)化工學(xué)院、生物學(xué)院教授,湖南大學(xué)2011分子工程與診療學(xué)協(xié)同創(chuàng)新中心主任,兼任佛羅里達(dá)大學(xué)杰出教授。2000年國(guó)家杰出青年基金B(yǎng)獲得者、2010年國(guó)家973重大科學(xué)研究計(jì)劃(蛋白質(zhì)組)首席科學(xué)家,2012年國(guó)家自然基金會(huì)創(chuàng)新群體負(fù)責(zé)人,2013年度“高等學(xué)校學(xué)科創(chuàng)新引智計(jì)劃”國(guó)家化學(xué)生物學(xué)創(chuàng)新引智基地(“111計(jì)劃”)帶頭人。擔(dān)任國(guó)家自然科學(xué)基金委化學(xué)部咨詢委員會(huì)委員,科技部重大研究計(jì)劃專家組成員等。曾任美國(guó)《Anal Chem》雜志副主編, 現(xiàn)任由中國(guó)化學(xué)會(huì)和英國(guó)皇家化學(xué)會(huì)共同主辦的“分析化學(xué)前沿”雜志主編, 以及JACS, ACS Nano、Chemical Science、中國(guó)科學(xué)化學(xué),國(guó)家科學(xué)評(píng)論等國(guó)內(nèi)外雜志編委。 譚蔚泓教授在Science、PNAS、J. Am. Chem. Soc.、Angew. Chem. Int.
